Abstract
The aim of this study was to determine whether agouti gene exon 2 was present in porcine. The approach could then be used for further identification of exons 3 and 4 and the cloning of the porcine agouti gene. Using PCR and Southern hybridization, one DNA fragment, PorAex2, was amplified from porcine (Yorkshire) liver DNA, it was 161 bp in length, the same size as human and mouse agouti exon 2. The PorAex2 has 79% and 75.5% nucleotide homology to human and mouse agouti exon 2, respectively. The predicted amino acid sequence of PorAex2 has 85% and 74% homology to human and mouse agouti exon 2, respectively. These results suggest that swine have the agouti gene and at least exon 2 of this gene is similar to that of humans and mice.
